Composition, linkage and side-chain modification analysis of sialic acids, including O-acetylation.
- Composition- Sialic acids (N-acetylneuraminic acid and N-glycolylneuraminic acid) composition from mammalian, and bacteria glycoproteins are analyzed by High Performance Anion Exchange Chromatography with Pulsed Amperometric Detection (HPAEC-PAD)
- Linkage– Tandem MS fragmentation can be used to quickly determine linkage 2,3 vs 2,6 linked sialic acid based on diagnostic fragmentation ions typical to each linkage. Critical data regarding the sialylation of an engineered or isolated glycoforms is obtained. Alternatively, enzymatic method for determining 2,3 vs 2,6 linked sialic acid may be used in conjunction with HPAEC-PAD monosaccharide analysis.
