Glycoanalytics services

The BioF:GREAT User Facility, operating within the CCRC Analytical Services and Training Laboratory, provides carbohydrate and glycoconjugate analysis to academic, government and industrial users. Services run from monosaccharide composition through site-specific glycoproteomics, using HPAEC, GC-MS, MALDI-TOF/TOF, Orbitrap MS and NMR.

Available services

N- and O-linked glycomics

Profiling of released N- and O-linked glycans from purified glycoproteins, cells, tissues, culture supernatants and gel bands by MALDI-TOF/TOF and Orbitrap mass spectrometry.

Glycoproteomics

Site-specific characterisation of protein glycosylation by protease digestion and ETD/HCD tandem mass spectrometry.

Glycolipids and additional analyses

Extraction and characterisation of glycosphingolipids and glycosylphosphatidylinositols, plus sulfation/phosphorylation analysis, SDS-PAGE, in-gel digestion and lectin blotting.

Glycan sequencing by tandem MS

Sequence determination of released glycans by tandem mass spectrometry.

Monosaccharide and linkage analysis

Monosaccharide composition by HPAEC and GC-MS, glycosyl linkage analysis by methylation GC-MS, and exoglycosidase digestion.

Sialic acid analysis

Composition, linkage and side-chain modification analysis of sialic acids, including O-acetylation.

Sites of glycosylation

N- and O-linked site mapping, including 18O-labelling and LC-MS isotopic shift monitoring.

How to request a service

  1. Identify the analysis. Select the service area above; each page lists the methods used, the information returned and typical sample requirements.
  2. Submit a request. Complete the service request form describing the sample, its origin and the question you need answered.
  3. Consultation. Facility staff confirm the analytical approach, sample amount, turnaround time and cost.
  4. Sample submission. Ship the sample with the agreed documentation.
  5. Report. You receive processed data with an interpretation, and follow-up consultation where the result warrants it.
Larger projects. Requests that go beyond routine analysis — method development, large sample series or collaborative research — are handled through the user proposal route rather than the service request form. If you are unsure which applies, see the user flow chart.
